The mitotic spindle has a crucial role in ensuring the accurate segregation of chromosomes into the two daughter cells during cell division, which is paramount for maintaining genome integrity. It When a cell starts to divide, it forms a spindle, a micro-machine made of microtubules, which separates the duplicated chromosomes. The attachment of microtubules to chromosomes is mediated by kinetochores, protein complexes on the chromosome. We recently analysed a nationwide cohort of 113 491 sera consecutively tested for ANA which was positive in 60 501 (53%).2 Of these, 592 samples with staining for anti-mitotic spindle apparatus (MSA) antigen patterns (NuMA/MSA-1, midbody/MSA-2, CENP-F/MSA-3 and centrosome) were chosen for further analysis.

Mitotic spindle

2). muscle spindle a mechanoreceptor found between the skeletal muscle fibers; the muscle spindles are arranged in parallel with muscle fibers, and respond to passive stretch of the muscle but cease to discharge if the muscle contracts isotonically, thus signaling muscle length. The mitotic spindle is the macromolecular machine that segregates chromosomes to two daughter cells during mitosis. The major structural elements of the spindle are microtubule polymers, whose intrinsic polarity and dynamic properties are critical for bipolar spindle organization and function.

Thus, when the cell divides, both daughter cells have complete… Mitotic spindle. The mitotic spindle is a transient microtubule -based structure that is present during mitosis and that mediates accurate separation of replicated sister chromatids to the daughter cells. Upon entry into mitosis, the interphase network of microtubules disassembles, and the duplicated centrosomes move to opposite ends of the cell.

When a cell starts to divide, it forms a spindle, a micro-machine made of microtubules, which separates the duplicated chromosomes. The attachment of microtubules to chromosomes is mediated by kinetochores, protein complexes on the chromosome. The spindle is the structure that pulls the chromatids to opposite ends of the cell during the cell division processes of mitosis and meiosis. Once separated, each chromatid becomes a chromosome.

To investigate its role during spindle formation, we tested spindle assembly in frog egg extracts from which NuMA was immunodepleted. Immunodepletion revealed that NuMA forms a complex with cytoplasmic dynein and dynactin. This volume includes a series of protocols focused on mitotic spindle assembly and function. The methods covered in this book feature a broad range of techniques from basic microscopy to the study of spindle physiologies relevant to cancer. The mitotic spindle is a chiral object with left-handed helicity of microtubule bundles.
